肾炎四味片可减轻5/6肾切除大鼠的肾脏纤维化

［摘要］目的 观察肾炎四味片对5/6肾切除大鼠肾组织核转录因子-κB(NF-κB)、转化生长因子-β1(TGF-β1)表达的影响。方法 24只Wistar大鼠随机分成假手术组(A组)、模型组(B组)、干预组(C组)、空白对照组(D组)，C、D两组术后3天予肾炎四味片灌胃，术后第90天处死，Real Time PCR和western blot分别测定组织中NF-κB、TGF-β1的mRNA和蛋白表达水平。结果 与A组相比，B组的NF-κB、TGF-β1的mRNA和蛋白水平表达增强，具有统计学差异(P＜0.05)，C组虽然高于A组(P＜0.05)，但其表达低于B组，具有统计学差异(P＜0.05)；免疫组化结果类似。NF-κB、TGF-β1二者的表达存在密切关系。结论 肾炎四味片可能通过下调NF-κB、TGF-β1的mRNA和蛋白的表达发挥治疗肾脏纤维化的作用。

Shenyansiwei tablet could alleviate the fibrosis of 5/6 subtotal nephrectomized rats

［Abstract］ Objective To observed the expression of NF-κB and TGF-β1 by shenyansiweipian Tablet in 5/6 subtotal nephrectomized rats. Methods 24 male Wistar rats were randomly divided into 4 groups, sham-operated group(group A), subtotal nephrectomy(STN) group(group B), STN with shenyansiwei tablet treated group(group C), and nomal rats with shenyansiwei tablet treated group(group D) . These rats were intragastric administrated with shenyansiwei Tablet at 3 days after operation,and were sacrificed at 90 days after operation,analyzed the changes of expression of NF-κB and TGF-β1 by routine immunohistochemical method. Result Immunohistochemical results: The expression of NF-κB and TGF-β1 in group B gradually increased, and there was significantly difference in the same group at the three time points, and that in group C was significantly higher compared with group A of the same stage(P＜0.05),and significantly lower compared with group B (P＜0.05).There was positive correlation between the two cytokines of NF-κB and TGF-β1, and there was positive correlation between the two cytokines metioned above and serum creatinine.
